ABSTRACT

@#AIM: To evaluate the effect on accommodative functions after femtosecond laser assisted LASIK(FEMTO LASIK)for myopic anisometropia.<p>METHODS: A respective serial case observational study was designed. Thirty-five patients(70 eyes)with myopic anisometropia treated by FEMTO LASIK were included in the study. Their uncorrected distance visual acuity, distance spherical equivalent, the difference of binocular diopter and accommodative functions including amplitude of accommodation, negative relative accommodation, positive relative accommodation and lag of accommodation were measured three times(before surgery, 1mo and 3mo after surgery). <p>RESULTS: At 1mo and 3mo after surgery, uncorrected distance visual acuity was improved significantly, distance spherical equivalent and the difference of binocular diopter were significantly decreased. Amplitude of accommodation and positive relative accommodation were improved at 3mo after surgery. The differences were statistically significant. There was no statistically significant difference of negative relative accommodation and lag of accommodation between these three times.<p>CONCLUSION: FEMTO LASIK for the correction of myopic anisometropia is an effective and safe procedure. The surgery may have no adverse influence on patients' accommodative function. It would be helpful for recovery of the binocular coordinate accommodative function.

ABSTRACT

AIM:To explore the effects of genipin (GEN) on high glucose (HG) -induced oxidative stress injury and apoptosis in H9c2 cardiomyocytes.METHODS:H9c2 cells were cultured in vitro and HG-induced injury model was established.H9c2 cells were divided into 4 groups:normal control (NC) group (glucose at 5.6 mmol/L) , HG group (glucose at 50 mmol/L) , NG+GEN group and HG+GEN group.The concentration of genipin was used at 10μmol/L.The viability of the H9c2 cells was measured by CCK-8 assay.The intracellular malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were determined by enzyme labeling and WST-1 methods, respectively.The activity of lactate dehydrogenase (LDH) in the cell culture supernatant was detected by microplate method.Fluorescent probe DCF was used to detect intracellular levels of reactive oxygen species (ROS).Nucleosome fragments was measured to evaluate cell apoptosis by ELISA.The intracellular mitochondrial membrane potential was detected by JC-1 method.The protein levels of Mn-SOD, cytochrome C (Cyt C) , Bax and cleaved caspase-3 were determined by Western blot.RESULTS:Compared with HG group, the cell viability in HG+GEN group was increased significantly (P<0.05) , the levels of MDA and LDH were decreased (P<0.05) , SOD activity was increased (P<0.05) , the levels of ROS and nucleosome fragments in HG+GEN group were decreased (P<0.05) , and the mitochondrial membranes potential was notably increased (P<0.05).Compared with NG group, the activation of Mn-SOD was decreased, but the protein levels of Cyt C, Bax and cleaved caspase-3 were increased in HG group (P<0.05).Compared with HG group, the activation of Mn-SOD was increased, and the protein levels of Cyt C, Bax and cleaved caspase-3 were decreased in HG+GEN group (P<0.05).CONCLUSION:Genipin protects HG-induced H9c2 cardiomyocytes against oxidative stress injury and apoptosis.

ABSTRACT

@#AIM: To research the protection of Mcc950, the inhibitor of NLRP3, against the inflammatory injury to human retinal pigment epithelium cell line ARPE-19. <p>METHODS: Cultured cell line ARPE-19 was divided into control group, H<sub>2</sub>O<sub>2</sub> treating group, Mcc950 treating group and Mcc950+H<sub>2</sub>O<sub>2</sub> treating group. Different concentrations of H<sub>2</sub>O<sub>2</sub> and Mcc950 were used to treat the cells. Cell activity was detected by using CKK8 and proper experimental concentration of H<sub>2</sub>O<sub>2</sub> and Mcc950 were determined. After the treatment, the concentration of IL-1β were detected by using ELISA. The change of NLRP3 related proteins were detected by Western blot. And cell apoptosis was examined by TUNEL stain. <p>RESULTS: Cell ability was gradually decreased along with the increasing treating concentrations of H<sub>2</sub>O<sub>2</sub>. Cell ability showed statistical difference when the concentration of H<sub>2</sub>O<sub>2</sub> arrived 400μmol/L. With the concentration of 0.1 and 1μmol/L, Mcc950 had no effect on cell ability. So we chose 400μmol/L H<sub>2</sub>O<sub>2</sub> and 1μmol/L Mcc950 as the experimental concentrations. Compared with the normal control group, the cell viability in the H<sub>2</sub>O<sub>2</sub> treating group was significantly reduced, the IL-1β in the supernatant was significantly increased, and the apoptosis rate was significantly increased, with statistically significant differences(<i>P</i><0.05). In Mcc950+H<sub>2</sub>O<sub>2</sub> treating group, cell viability was significantly increased, the IL-1β in the supernatant and the apoptosis rate were significantly decreased(<i>P</i><0.05). By Western blot, after treated with 400μmol/L H<sub>2</sub>O<sub>2</sub>, the IL-1β, NLRP3, pro-caspase1 and caspase1 were obviously increased compared to control group. After treated with Mcc950, the NLRP3 and pro-caspase1 still were at high level, the expression of caspase1 was suppressed, which indicated that Mcc950 effectively inhibited the activation of NLRP3 inflammasome consequently disturbed the formation of caspase1.<p>CONCLUSION: Mcc950 can inhibits the function of NLRP3, leading to increasing of the cell ability and decreasing of the cell apoptosis.

ABSTRACT

Objective: To compare therapeutic effect and safety of dabigatran and warfarin anticoagulant therapy during catheter ablation in patients with atrial fibrillation (AF). Methods: Clinical data of 325 AF patients, who received catheter ablation in our hospital from Jan 2011 to Aug 2014, were retrospectively analyzed. According to perioperative anticoagulant therapy plan, patients were divided into dabigatran group (n=187) and warfarin group (n=138). General data, international normalized ratio (INR), activated coagulation time (ACT), success rate of pulmonary vein isolation (PVI), operation time and incidence rate of complications were compared between two groups. Results: Compared with warfarin group, there were significant reductions in percentages of diabetes mellitus (20. 3% vs. 11. 8%), chronic heart failure (19. 6% vs. 10. 2%) and left atrial diameter [(47±10) mm vs. (44± 9) mm]in dabigatran group, P<0. 05 or<0. 01. INR of dabigatran group was significantly lower than that of warfarin group [(1. 3±0. 3) vs. (2. 4±0. 4), P=0. 001], and there were no significant difference in ACT, success rate of PVI and operation time between two groups (P>0. 05 all). Incidence rate of main complications in dabigatran group was significantly lower than that of warfarin group (0. 5% vs. 5. 1%, P=0. 025). Conclusion: Compared with warfarin, there is significant reduction in INR and incidence rate of main complications during catheter ablation in dabigatran group, which is worth extending.

ABSTRACT

AIM:To observe the effects of ginsenoside Rh 1 on the levels of inflammatory factors in serum and bronchoalveolar lavage fluid(BALF),and the pathological changes of the lung tissues in an experimentally induced mouse asthma model.METHODS:Male BALB/c mice(n=40)were divided into 4 groups:normal control group,asthma mo-del group,and low-dose(40 mg· kg-1 · d-1 )and high-dose(80 mg· kg-1 · d-1 )ginsenoside Rh1 groups.The bron-chial asthma mouse model was established by the method of ovalbumin induction and excitation ,and during the excitation period,the mice were daily treated with ginsenoside Rh 1 for 2 weeks.At 24 h after the final dose of ginsenoside Rh 1,the mice were sacrificed.The number of eosinophils(EOS)and the concentrations of interleukin(IL)-4,IL-5 and interferon(IFN)-γin BALF were determined.The levels of IgG and IgE in serum were measured ,and the expression of transforming growth factor(TGF)-β1 and the pathological changes in lung tissues were evaluated.RESULTS:Ginsenoside Rh1 inhibi-ted the increases in the number of EOS and the concentrations of IL-4,IL-5,IFN-γand IgE,reversed the increased ex-pression of TGF-β1,and improved the pathological changes of the lung tissues in asthmatic mice.CONCLUSION:Gin-senoside Rh1 improves the immuno-inflammatory profile and pathological changes in the experimentally induced mouse asth -ma model,implying its potential therapeutic effect on asthma.

ABSTRACT

AIM: To evaluate the effect of orthokeratology on progression of juvenile myopia and analysis its influencing factors.METHODS: Totally 97 patients (189 eyes,aging from 8 to 17 years old) who received orthokeratology lenses treatment in our hospital from January 2012 to December 2014,were followed up for 2a.The visual acuity,corneal curvature,diopter,and ocular axial length were observed.Factors of influencing myopia progress in juvenile were analyzed.RESULTS: At 1mo after receiving orthokeratology contact lenses,the visual acuity and corneal curvature were changed compared with that of before(P<0.001).After 2a of receiving orthokeratology contact lenses,the difference was significant compared with baseline: spherical equivalence (-0.51±0.64D,t=10.864,P<0.001),axial length(0.33±0.31mm,t=14.879,P<0.001),corneal astigmatism (-0.25±0.43D,t=5.375,P<0.001).Statistic analysis showed that there was a negative correlation between the spherical equivalence and age,baseline of diopter or ocular axial length(P<0.05).CONCLUSION: Orthokeratology can effectively improve the visual acuity of patients.Although there is slightly progression in diopter and ocular length after 2a of wearing orthokeratology contact lenses.Orthokeratology is an effective treatment on controlling progression of juvenile myopia,especially in the elder children who with the longer basic axial length and the greater diopter.

AIM:To observe the effects of Dickkopf-3 ( Dkk-3 ) in diabetic retinopathy ( DR ) circulating blood in patients with the expression level, the Dkk - 3 development changes in the diabetic retinopathy of significance in the diagnosis of early DR. ● METHODS: Eighty - five type 2 diabetic patients, included the non - proliferative diabetic retinopathy ( NPDR ) 23 patients, proliferative diabetic retinopathy, proliferative DR ( PDR ) in patients with 30 and non-diabetic retinopathy ( NDR ) with 32 cases. The same period of healthy physical examination was selected as control group ( 80 cases ) . Serum samples were collected, and the relative expression level of Dkk-3 was detected by enzyme - linked immunosorbent assay ( ELlSA) double antibody sandwich assay. The statistical differences were compared between groups. ●RESULTS: The plasma level of Dkk - 3 ( 430. 16 ± 198. 11pg/mL) in DR patients was significantly lower than that in healthy control group (627. 48±294. 45 pg/mL; P ●CONCLUSION:The decrease of Dkk-3 level may be related to the occurrence and development of diabetic retinopathy, and there is a significant correlation with PDR. Circulating blood Dkk - 3 protein in diabetic retinopathy has a certain differential efficacy, it is likely to become diabetic retinopathy patients peripheral blood test indicators.

ABSTRACT

?AlM:To investigate the sensitive parameters of the anterior chamber changes with Pentacam anterior segment analysis system before and after laser peripheral iridectomy (LPl) in primary angle-closure suspetive (PACS). ? METHODS: Sixty eyes of 33 PACS patients were enrolled in this study. Pentacam examination was performed before and 1d after LPl to measure the central anterior chamber depth ( CACD ) , the peripheral anterior chamber depth ( PACD ) , the anterior chamber volume ( ACV) and the peripheral anterior chamber angle ( ACA) . Statistical analysis used paired t test. ?RESULTS: There was no statistical significance on the changes of ACD. PACD and ACV increased significantly between before and 1d after LPl. ACA was widened from (22. 26o±5. 18o) to (26. 42o±5. 20o), which were increased significantly between before and 1d after LPl. ?CONCLUSlON: LPl can deepen the PACD and increase the ACV in PACS. PACD and ACV are the sensitive parameters of the anterior chamber changes with Pentacam anterior segment analysis system.

ABSTRACT

Background Posterior capsular opacification (PCO)affects the pseudoaccommodation of 1CU accommodative intraocular lens (1CU AIOL).At present,few studies on the effect of PCO and Nd∶ YAG laser capsulotomy on intraocular shifting of 1CU AIOL are published.Objective The present study was to evaluate the effect of PCO and Nd∶YAG laser capsulotomy on the shifting of 1CU AIOL.Methods A respective serial caseobservational study was designed.Written informed consent was obtained from each patient prior to this study.Twentyfour eyes of 20 patients with PCO after phacoemulsification and implantation of 1CU AIOL were included in this study.Ocular examination was performed 3 months after IOL implantation,1 day before Nd:YAG laser capsulotomy and 3 months after Nd∶YAG laser capsulotomy to evaluate the distance corrected near visual acuity(DCNVA).The difference in the anterior chamber depths before and after administering 1% pilocarpine topical eye drops was measured with the IOLMaster to determine the intraocular shifts of the IOL.The extent of IOL shifting was compared among 3 time points to assess the factors influencing IOL accommodation after 1CU AIOL implantation.Results The shifting amplitude of 1CU AIOL was(0.44±0.21)mm 3 months after implantation of 1CU AIOL,(0.27±0.11)mm 1 day before Nd ∶ YAG laser capsulotomy,and (0.34±0.10) mm 3 months after Nd ∶ YAG laser capsulotomy,showing a significant difference among them(F=7.180,P=0.001).The shifting amplitude of 1CU AIOL significantly declined 1 day before Nd∶YAG laser capsulotomy in comparison with 3 months after implantation of 1 CU AIOL(P =0.006).The shifting amplitude 3 months after Nd∶YAG laser capsulotomy increased slightly in comparison with 1 day before Nd∶YAG laser capsulotomy(P=0.059).DCNVA was(3.1±0.9)J 3 months after implantation of 1CU AIOL,(6.2±0.8) J 1 day before Nd ∶ YAG laser capsulotomy and(3.4±0.7) J 3 months after Nd ∶ YAG laser capsulotomy,with a significant difference among them (F =110.270,P =0.000).DCNVA was lower 1 day before Nd∶ YAG laser capsulotomy than 3 months after implantation of 1CU AIOL(P<0.05).However,DCNVA was higher 3 months after Nd∶YAG laser capsulotomy than that of 1 day before Nd∶YAG laser capsulotomy (P<0.05).There was no significant correlations between DCNVA and IOL movement 3 months after IOL implantation,1 day before Nd∶ YAG laser capsulotomy and 3 months after Nd ∶ YAG laser capsulotomy (r1 =-0.150,P1 =0.486,r2 =-0.320,P2 =0.122,r3 =-0.100,P3 =0.633).Conclusions The shifting amplitude of 1CU AIOL markedly declines due to PCO.No clinically significant influence of Nd ∶ YAG laser capsulotomy on the shifting amplitude of 1 CU AIOL is found.DCNVA can improve after Nd∶YAG laser capsulotomy.Multiple inter-related factors concerning pseudophakic accommodation may influence DCNVA.

ABSTRACT

<p><b>OBJECTIVE</b>To obtain the prevalence of allergic rhinitis (AR) in different regions of northern China, to analyze the correlation and interaction between AR and bronchial asthma (BA) or atopic dermatitis (AD), and to provide reference for the prevention and treatment of allergic diseases such as AR.</p><p><b>METHODS</b>To obtain the indexes including age, occupation, atopic physical fitness, smoking, alcohol, lifestyle and so on. To explore the correlation between AR and BA or AD. From April 2007 to May 2010, the serum specific IgE (sIgE) was investigated in different regions (rural areas of Qingxian, Hebei; coastal fishing village of Bohai Bay, Huanghua; area of Wuling Mountain, Chengde; urban areas of Tianjin) by randomly multi-stage and cluster sampling, with total population of 1524.</p><p><b>RESULTS</b>The prevalence of AR, BA and AD were 9.1%, 5.4% and 6.0%. The prevalence of BA and AD were 30.9% and 29.5% in AR patients, but were 2.9% and 3.7% in non-AR, and the differences were of statistical significance (χ(2) values were 192.97 and 148.40, respectively, all P < 0.01). The risk of people with BA suffering from AR was as 8.619 times as those free from BA, the risk of people with AD suffering from AR was as 1.817 times as those free from AD and, the risk of workers suffering from AR was as 2.320 times as farmers in terms of working factor by analysis of Logistic regression for AR and BA, AD, age, occupation, atopic physical fitness and other factors.</p><p><b>CONCLUSIONS</b>The prevalence of AR are correlated with BA and AD. It is greater between BA and AR than AD and AR in the strength of correlation.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To obtain the prevalence and related factors in allergic rhinitis (AR) and other allergic diseases in rural area in China through epidemiological investigation with large sample and multi-faceted survey data.</p><p><b>METHODS</b>Face to face survey was conducted in different regions (rural areas of Cangzhou, Hebei, coastal fishing village of Bohai Bay, area of Wuling Mountain, Chengde, urban areas of Tianjin) from April 2007 to May 2009. In the same time, serum specific IgE (sIgE) was detected in the digits of every 0, 1or 5 in them. SPSS 13.0 software was used to analyze the data.</p><p><b>RESULTS</b>Five thousand and ten cases were investigated. There were 823 cases with the symptoms or signs of AR (16.4%). Four hundred and two cases were found to have positive serum sIgE antibody in 1576 detected cases (25.5%). One hundred and fourty-six cases with nasal allergic symptoms or signs were diagnosed as AR. The incidence of AR was 9.3% (146/1576). The occurrence of allergic symptoms or signs had a significant statistical difference with factors such as age, occupation, atopic constitution (χ(2) value were 7.96, 9.73, 16.53, 8.95 respectively, all P < 0.05), and it was closely related to lower respiratory tract allergies (β = 2.544, P < 0.01). The most common allergens were dog and cat epithelium in rural areas and dust mites in city.</p><p><b>CONCLUSIONS</b>The incidence of AR is higher whether in urban or rural areas, it should be taken seriously as the impact on human health. The occurrence is closely related to physical characteristics and environmental factors.</p>
